What Is the Role of Toll-Like Receptors in Lynch Syndrome?

This presentation by Dr. Deanna Windham explores the critical role of Toll-like Receptors (TLRs) in immune responses and their complex involvement in Lynch syndrome.  It highlights how TLR dysregulation can promote tumor initiation and progression, particularly in colorectal cancer, and discusses the potential of TLR-targeting immunotherapies.

Dr. Deanna Windham, DO:  Some of the other factors around Lynch syndrome other than epigenetics and genetics is toll-like receptors which play a crucial role in the immune response by detecting pathogens typically um and triggering the production of inflammatory cytoines. But in lich syndrome the toll-like receptors are um h either hyperactivated or hypoactivated. Either way, it is whether it is hyper or hypoactivated, it's having a negative impact in cancer and antagonizing these TLRs uh receptors are one of the promising imunotherapy approaches to cancer coming up. Um and so this research was talking about toll-like receptors specifically to Lynch syndrome and suggested that disregulation of TLRs can contribute to the chronic inflammation and immune dysfunction observed in Lynch syndrome and that this may in turn promote tumor initiation and progression. These researchers found that uh specifically in colarctal cancer in lynch syndrome patients um they theorized that activation of pro-inflammatory pathways was implicated in the development of the disease and while the precise mechanisms by which TLRs influence the development and progression of cancer and lynch syndrome are not fully understood it is understood that it does uh have an impact and play a part.
